چکیده
Using Biochemica Test Kits (Boehringer GmbH, Mannheim) a significant difference in alanine aminotransferase : aspartate aminotransferase (ALAT : ASAT) ratios was found between Trypanosoma brucei brucei (4 strains; mean ± S.E.; 4.204 ± 0.342) and T. b. rhodesiense (4 strains; mean ± S.E.: 2.973 ± 0.174) blood¬ stream stage crude soluble enzyme extracts. This difference is a stable characteristic, unaffected by the strains tested, level of parasitaemia, and number of subpassages in rats. There was also a significant difference between T. b. brucei and T. b. rhodesiense specific ALAT activities. ALAT : ASAT ratios and specific ALAT activities in bloodstream and culture slages of T. b. brucei were significantly different. Control tests elucidated the value of the test assay used for crude soluble enzyme preparations of both T. brucei subspecies. Within either subspecies there was a positive significant correlation between the degree of pleomorphism (percentage of slender forms) and ALAT : ASAT ratios.

Methionine regeneration and aspartate aminotransferase in parasitic protozoa.
Aspartate aminotransferases have been cloned and expressed from Crithidia fasciculata, Trypanosoma brucei brucei, Giardia intestinalis, and Plasmodium falciparum and have been found to play a role in the final step of methionine regeneration from methylthioadenosine. All five enzymes contain sequence motifs consistent with membership in the Ia subfamily of aminotransferases; the crithidial and ...
متن کاملEfficacy of repeated doses of diminazene aceturate (Dinazene®) in the treatment of experimental Trypanosoma brucei infection of Albino rats
The efficacy of repeated doses of Dinazene® in Albino rats experimentally infected with Trypanosoma brucei (Gboko strain) was investigated. A total of 30 adult female Albino rats weighing 130-190 g were used for the study. They were assigned to six groups (groups A-F) of five rats each. Groups A-D were infected intraperitoneally with 1.0 × 106 trypanosomes in 400 μL of PBS diluted blood while g...
متن کاملKnockdown of Asparagine Synthetase A Renders Trypanosoma brucei Auxotrophic to Asparagine
Asparagine synthetase (AS) catalyzes the ATP-dependent conversion of aspartate into asparagine using ammonia or glutamine as nitrogen source. There are two distinct types of AS, asparagine synthetase A (AS-A), known as strictly ammonia-dependent, and asparagine synthetase B (AS-B), which can use either ammonia or glutamine. The absence of AS-A in humans, and its presence in trypanosomes, sugges...
متن کاملTargeting of cytoskeletal proteins to the flagellum of Trypanosoma brucei.
The eukaryotic flagellum represents one of the most complex macromolecular structures found in any organism and contains more than 250 proteins. Due to the relative ease of genetic manipulation the flagellum of Trypanosoma brucei has emerged as an accessible model system to study the morphogenesis and dynamics of this organelle. We have recently started to characterise the mechanisms by which c...
متن کاملTranscript Expression Analysis of Putative Trypanosoma brucei GPI-Anchored Surface Proteins during Development in the Tsetse and Mammalian Hosts
Human African Trypanosomiasis is a devastating disease caused by the parasite Trypanosoma brucei. Trypanosomes live extracellularly in both the tsetse fly and the mammal. Trypanosome surface proteins can directly interact with the host environment, allowing parasites to effectively establish and maintain infections. Glycosylphosphatidylinositol (GPI) anchoring is a common posttranslational modi...
